Notes: The Invasive Breast Cancer Profile includes immunohistochemical staining and quantitative image analysis of ER, PR, Her2/neu, and Ki-67.
Clinical Utility: The Invasive Breast Cancer Profile is used to assess patients with invasive breast carcinoma for treatment and prognosis. ER and PR are used to assess tumor response to endocrine therapy. Her2/neu by IHC can be used to assess patient's eligibility for treatment with Herceptin. A reflex to Her2/neu by FISH is performed on all 2+, 3+, and triple negative carcinomas (ER, PR and Her2/neu negative).

Unacceptable Conditions: Specimens fixed using a fixative other than formalin are not acceptable for testing. If unstained slides are provided for testing, slides received more than 6 weeks after cutting are not acceptable for IHC testing.

Formalin fixed paraffin embedded tissue with representative tumor.

A tissue block or 10, 4µ thick, unstained, positively charged slides are acceptable for testing (this includes slides needed for a reflex to Her2/neu FISH).

The cold ischemic time (time of removal from the body to the time in formalin) should be ≤ 1 hour. The time in formalin (fixation) should be 6-48 hours. Specimens should not be decalcified with strong acids.
